S-One Trio is a prescription medication used to treat advanced gastric cancer, including adenocarcinoma of the stomach, and gastroesophageal junction (GEJ) adenocarcinoma.
Composition:
S-One Trio is a combination of three active ingredients:
- Gimeracil (5.8mg): a dihydropyrimidine dehydrogenase (DPD) inhibitor
- Oteracil (15.8mg): an inhibitor of the enzyme orotidine 5′-monophosphate decarboxylase
- Tegafur (20mg): a pro-drug of fluorouracil (5-FU)
Mechanism of Action:
S-One Trio works by:
- Inhibiting the enzyme DPD, which helps to increase the levels of 5-FU in the body
- Inhibiting the enzyme orotidine 5′-monophosphate decarboxylase, which helps to increase the levels of 5-FU in the body
- Releasing 5-FU, which is then converted into fluorouracil (5-FU) in the body
- 5-FU is then metabolized into fluorouridine triphosphate (FUTP), which inhibits the synthesis of DNA, RNA, and protein
Uses:
S-One Trio is used to treat:
- Advanced gastric cancer, including adenocarcinoma of the stomach, and gastroesophageal junction (GEJ) adenocarcinoma
- Patients who have not received prior chemotherapy for gastric cancer
Dosage:
The recommended dosage of S-One Trio is:
- 1 tablet taken orally twice daily, with or without food
- Continue treatment until disease progression or unacceptable toxicity

- Fatigue
- Nausea and vomiting
- Diarrhea
- Stomach pain
- Decreased appetite
- Increased risk of infection
- Increased risk of bleeding
- Low white blood cell count
- Low platelet count
